2. Put the disinfection vaginal probe into the vagina, detect the ovary and mature follicles in the fornix, fix the probe on the puncture site, make the mature follicles located on the puncture guidance line, and measure the needle depth.

3. Under the guidance of vaginal ultrasound, the needle pierces the vagina and fornix to the follicle. When the needle reaches the surface of the follicle, after a short pause, moderate force and acceleration should be applied to rapidly penetrate the needle into the follicle cavity.

4. Confirm that the tip is located in the follicular cavity, turn on the negative pressure suction device, and aspirate the follicular fluid until the follicle collapses. After draining the follicle fluid from one follicle, the needle is inserted into the adjacent larger follicle. If the number of dominant follicles is less than 4~5, or there are more immature follicles, the double-cavity oocyte needle should be used. If necessary, the follicular cavity can be washed several times with culture medium and repeated aspiration 2 ~ 3 times.

If the number of dominant follicles is more than 4~5, single cavity oocyte needle can be used. If the number of oocytes taken out is less than the number of follicles observed under ultrasound during oocyte collection, double cavity oocyte needle should be used. Each follicle should be rinsed 2 ~ 3 times with culture fluid in order to obtain more oocytes and avoid loss.

5. Puncture all follicles with a diameter greater than 10mm as far as possible, and the follicles located on the same puncture line can be completed from shallow to deeper than one time into the needle. For follicles on different puncture lines, the needle was withdrawn to the ovarian surface without exiting the vaginal wall, and then the puncture direction was changed. After the end of one puncture, the other ovarian puncture was performed.

6. At the end of puncture, withdraw the puncture needle out of the body. Routine pelvic scan for possible internal bleeding or hematoma formation.
